Screening of extremozyme production of selected extremophilic PHA producers

This bachelor thesis deals with the screening of the production of extracellular hydrolytic enzymes in thermophilic microorganisms of the genera Aneurinibacillus, Brevibacillus, Chelatococcus, Pseudomonas, Schlegelella, Tepidimonas and Caldimonas. The ability of selected enzymes, namely proteases, lipases, amylases, xylanases, cellulases and pectinases, was tested in the investigated microorganisms. Such testing made it possible to assess in which microorganisms the production of specific enzymes can be observed. Based on the results of the screening, it was found that Schlegelella aquatica LMG 23380, Tepidimonas fonticaldi LMG 26746 and the investigated microorganisms of the genus Chelatococcus did not show the ability to produce any of the tested enzymes extracellularly. In natural isolates of Brevibacillus borstelensis LK 99 and Aneurinibacillus thermoaerophilus LK 102, only the ability to produce lipolytic enzymes was detected. The isolate Brevibacillus borstelensis Bz acts as a universal producer of all selected extremozymes. Enzyme activity was determined for selected producers. The bacterium Brevibacillus borstelensis Bz proved the ability to produce all six selected hydrolytic enzymes and has the highest activity of lipases, xylanases, cellulases and pectinases from the tested microorganisms. The highest proteolytic activity was measured in Thermomonas hydrothermalis DSM 14834 when cultured on skimmed milk powder.

Point mutation altering arginin for tryptophan amino acid residue in P1 site of tick salivary serpin Iripin-1 was created using specific primers. Recombinant protein with this mutation in nucleotide sequence was then expressed in chemically competent Escherichia coli cells, extracted from them and purified by affinity and size-exclusion chromatography. To see the impact of the mutation on inhibitory function of Iripin-1, its ability to bind trypsin and form covalent complexes was evaluated.
Selected proteolytic aspects as targets to combat ticks and tick borne pathogens

Ticks and tick-borne diseases (TBD) represent a growing global burden for both human and animal health. Tick-host-pathogen interactions have evolved through dynamic processes that accommodated the genetic traits of the hosts, pathogens transmitted, and the vector tick species that mediate their development and survival. As in other parasites, proteases and proteolysis have been found as one of the key factors in this interaction triangle. This thesis is focused on selected proteolytic aspects of tick and tick-borne diseases: (i) processing of host blood as a source of nutrients and energy (hematophagy) as a continuum of the long-term goal of the Laboratory of Vector Immunology, that established the currently accepted model of multienzyme degradation of host blood proteins by ticks (ii) proteases in innate immunity (iii) validation of Babesia proteasome as a potential therapeutic target against the tick transmitted apicomplexan parasites.
The role of NADPH oxidase and ros in invadopodia formation

Invadopodia as specific organelles enabling tumour cells movement, spreading over the organism and ultimately formation of metastasis are possible and promising targets of tumour therapy. Recently, many interesting facts about assembly and mechanism of function of invadopodia were discovered. Invadopodia are centres of ECM degradation by extra-cellular proteases facilitating an invasion of tumour cells. For creation of invadopodia a precisely localized increased production of ROS is necessary. ROS work as crucial signalling molecules and participate in many processes resulting in invadopodia formation. ROS in tumour cells are produced by specific extra-mitochondrial NADPH oxidases (Nox). Several regulatory molecules participating in activation and localization of Nox to invadopodia have been discovered recently (Tks organizer proteins). Furthermore, a regulatory role of Src kinase in ROS production and subsequent invadopodia formation was confirmed. The liming of forest soils and its implications for the forest ecosystem can be observed from many points of view. However, the assessment of liming effects may vary considerably. To evaluate the effect of liming on the mineral forms of nitrate and the activity of soil enzymes, proteases and ureases, the experimentally limed area was used. The liming was performed using finely milled lime dolomite in doses of 2, 3, 4, 6 and 26 t·ha-1. Mixed soil samples from the humification horizon H and the organomineral horizon Ah were taken from each variant. This mixed samples were further modified and subjected to laboratory analyses. The effect of liming on ammonium and nitrate nitrogen content and protease activity was found. In the case of NH4+, NO3-, soil proteases and ureases activity, differences were observed in relation to the change in the application dose of dolomitic limestone.
Možnosti snížení obsahu dusíkatých látek v krmných směsích pro brojlerová kuřata

The aim of the dissertation was to investigate the effect of exogenous protease (EP) in feed mixtures (FM) for broiler chickens with reduced crude protein (CP) content by 4 and 8 % on growth parameters, carcass quality, apparent ileal digestibility of amino acids and protease activity in the jejunum of broiler chickens. At the same time, the effect of EP on the given parameters was investigated at the increased level of rape seed meal (RSM) in FM. The addition of EP to KS with a 4% reduction in CP did not have a statistically significant (P> 0,05) effect on growth, conversion and feed intake, quality of carcass, nor on the apparent ileal digestibility of amino acids and protease activity in jejunum of the broiler chicken. Higher content of RSM in FM with a 4% CP reduction negatively (P <0.05) affected the protease activity in the jejunum. The addition of EP into FM with an 8% CP reduction statistically (P <0.05) increased the average live weight of broilers on the 17th and 24th day of the age. At a higher age, EP has not influenced on the live weight of broilers and the overall feed conversion. Females chickens consumed FM with 8% CP reduction had a significantly lower (P <0.05) weight of breast muscle and carcass value. Adding protease to FM with an 8% CP reduction statistically significantly (P <0.001) decreased the digestibility of Pro and Tyr (P <0.05). The EP had no significant (P> 0.05) effect on protease activity in the jejunum of broilers with FM intake with an 8% CP reduction. Higher content of RSM in FM with an 8% CP reduction significantly decreased (P <0.05) apparent ileal digestibility of Pro and significanlty (P <0.05) increased protease activity in broilers jejunum.
Phenotyping of proteolytic activities enabled by fluorogenic libraries

This work deals with the preparation of combinatorial libraries of peptides serving as platforms for proteolytic phenotyping. The primary objective was to prepare a solid phase fluorogenic peptide library and screen proteases by fluorescence. Further, the possibility of preparing solid phase DNA-encoded libraries was studied. Due to the non-reactivity of the specific proteases with the solid phase peptides, the solid phase was completely abandoned and DNA-encoded peptide library was prepared in the solution. Using this model of DNA-encoded dipeptide with terminal biotin, the new principle of testing proteolytic activities of proteases was verified. A combinatorial library of DNA-encoded hexapeptides was also prepared. Despite the low yield of the library, the possibility of DNA encoding, the amplifiability of the prepared molecules and the possibility of biotin-based separation were verified. The integrity of the hexapeptide sequence and the protease testing is the subject of further study.

The study of enzyme activities in compost

This thesis deals with the determination of enzyme activities in compost for consideration their changes during the composting. The changes in the activity of particular enzymes serve as the indicators of the activity of microorganisms, which are found in the compost. In the practical part of the work the changes in activity of proteases, cellulases, lipases, dehydrogenases and ureases within 21 days of composting under laboratory conditions were determined. The change in pH was observed as well. The changes in the pH-values and all enzyme activities except ureases present in the first week the same trend associated with the use of available substrates. The changes in the activities of the key enzymes in compost under laboratory conditions were compared with the changes in activities in compost under real conditions, which is the indicator of the different composting process under the given circumstances.
